It was developed to raise magnesium inside the brain, and in a 2010 rat study it did, along with better learning and memory in the rats. Why the two sleep trials disagree
Most articles cite the 2024 trial and stop there. The Australian trial, run by Clinical Research Australia in Perth with Murdoch University’s Adrian Lopresti as lead author, reached a more cautious conclusion. Three differences explain most of the gap.
- Who took part. The 2024 group was older, aged 35 to 55, and reported sleep problems. The Perth group was 18 to 45 and only dissatisfied with their sleep. People who sleep reasonably well have less room to improve. In the Perth trial, participants with more severe sleep problems did show fewer sleep disturbances than placebo.
- What was measured. Both used the same brand of sleep ring and got different answers on sleep stages. Consumer trackers estimate sleep stages rather than measure them the way a sleep lab does, so read the ring results in both trials with some caution.
- More was not better. Twice the dose for twice as long did not produce a bigger effect on ring-measured sleep. That argues against the escalating doses you see online.
Both trials were industry-funded. The 2024 trial was funded by AIDP, an ingredient company that employs three of its six authors. The Perth trial was funded by Threotech, which produces Magtein and supplied it for the study. That does not make the results wrong. It does mean no independently funded sleep trial exists yet.
The fair summary: magnesium L-threonate looks most useful for people who are over 35, already sleeping badly, and judging success by how they feel the next day. If you sleep reasonably well, the Perth results suggest you may notice changes in focus and heart rate variability before you notice anything in your sleep stages.

For context, the adult recommended daily intake of magnesium in Australia is 310mg to 420mg, depending on age and sex, and the upper limit from supplements is 350mg a day. No trial has compared the two head to head, so the choice comes down to what each has been tested for. Glycinate has its own sleep trial now. In a 2025 German study, 155 adults aged 18 to 65 who reported poor sleep took magnesium bisglycinate giving 250mg of elemental magnesium a day, or placebo. After four weeks the glycinate group’s sleep-difficulty scores had improved more than placebo, but the effect was small, and people with lower magnesium in their diets seemed to benefit most. There was no sleep ring or other objective measure.
That points to a practical split. Glycinate gives far more elemental magnesium per serve, 250mg in that trial against about 144mg in 2,000mg of threonate, so it suits people whose diets are low in magnesium. Threonate is the form studied for next-day function and focus, at a lower magnesium dose. If your sleep is disrupted by a busy mind and you care about daytime focus, threonate is the better match.
