Is 500mg enough?
On the trial evidence, 500mg is a reasonable middle dose. Four of the trials above found changes at 250mg, and every trial above that measured blood NAD found that it rose. The one trial that compared doses side by side found the largest rise in blood NAD at 600mg and 900mg, with 300mg also clearly above placebo (Yi et al., GeroScience, 2023). None of these trials used exactly 500mg, so the best we can say is that 500mg falls between doses that raised blood NAD.
What “enough” means is also unclear. Most of these trials measured blood NAD, walking tests or questionnaires over 4 to 12 weeks. A higher blood NAD reading has not been shown to slow ageing, and no trial has tested a dose over years. The dose-comparison trial was funded by two companies, Aba Chemicals and Abinopharm, and its lead author works for Abinopharm, as the paper declares. That does not cancel the result, but it is a reason to treat “600mg worked best” as one finding, not a settled rule.

Timing: when to take your daily 500mg
AgeMate’s directions put the scoop in the morning, and our NMN 500mg label also suggests the morning. That is a sensible habit, but the timing evidence is thin. The one trial designed to test timing split 108 older adults into morning and afternoon groups on 250mg a day for 12 weeks. The afternoon NMN group showed the largest improvements in a sit-to-stand test and in drowsiness, although some placebo groups improved too (Kim et al., Nutrients, 2022).

AgeMate’s label says to use it daily for up to 12 weeks unless a health professional advises otherwise. That lines up with the trials, which ran from 4 to 12 weeks, and with the lack of long-term data. - AgeMate’s label says adults only, notes that it contains phenylalanine and sulfites, and says it is not recommended for pregnant or breastfeeding women.
