What NMN does, and what the human trials show
NAD+ is used by many enzymes, including the sirtuins, and NMN is one step away from it in the body’s recycling pathway. The practical question is whether swallowing NMN raises NAD+ in people. Two trials say it does.
In a 2023 multicentre trial, 80 healthy middle-aged adults took a placebo or 300, 600 or 900 mg of NMN once a day for 60 days. Blood NAD rose in every NMN group, with the highest levels at 600 and 900 mg. Walking distance on a six-minute walk test improved more than with placebo, and no safety problems appeared. A measure of insulin resistance did not change (Yi et al., GeroScience). Three of the authors were employees of the chemical companies Abinopharm and Aba Chemicals, which is worth weighing.
In Japan, 42 healthy men aged 65 or older took 250 mg of NMN a day or a placebo for 12 weeks. NMN raised blood NAD+ and was well tolerated. Gait speed and left-hand grip showed improvements that the authors called nominally significant and in need of confirmation, and body composition did not change (Igarashi et al., npj Aging).
The wider picture is modest. A 2026 meta-analysis of 15 randomised trials, with doses from 250 to 2,000 mg a day for 14 days to 24 weeks, found no rise in adverse events and no significant change in liver enzymes. It also found no significant effect on body weight, fasting glucose, HbA1c, blood lipids or systolic blood pressure, with a slight fall in diastolic pressure (Yang et al., Nutrients). So NMN reliably lifts NAD, is well tolerated in the short term, and has not yet shown broad health effects in people. Nobody has shown that it extends human lifespan.

What resveratrol does, and what the human trials show
Resveratrol became famous for activating SIRT1 in the lab, but that finding has been disputed for years. A 2010 study found that resveratrol only appeared to activate SIRT1 when the test substrate carried a fluorescent tag, and not with natural substrates (Pacholec et al., Journal of Biological Chemistry). A 2013 study replied that activation does occur with substrates carrying certain water-repelling motifs, through a site on the enzyme it identified (Hubbard et al., Science). Both were lab experiments. Neither tells you what a capsule does in your cells.
Absorption is the next hurdle. When six volunteers swallowed 25 mg of labelled resveratrol, at least 70% was absorbed, but the gut and liver converted it so fast that only trace amounts circulated unchanged (Walle et al., Drug Metabolism and Disposition).
The human outcome data are small but real. In a German trial, 46 healthy overweight older adults took 200 mg of resveratrol a day or a placebo for 26 weeks. The resveratrol group retained more words over 30 minutes and showed stronger functional connectivity in the hippocampus, a brain region used for memory (Witte et al., Journal of Neuroscience). On the other side, a study following 783 Italians aged 65 and over for nine years found that the resveratrol they got from their diet was not linked with how long they lived (Semba et al., JAMA Internal Medicine). That study measured food intake, not supplements.

Do you need both NMN and resveratrol?
The reason for pairing them is mechanical. Sirtuins need NAD+ to work. NMN supplies NAD+, and resveratrol is proposed to switch SIRT1 on. A 2022 review describes exactly this split, with NMN and NR as substrates for NAD+ and resveratrol binding SIRT1 directly, and argues that combinations acting in complementary ways may have potential (DiNicolantonio et al., Open Heart). The only direct test we found was in mice. Mice given NMN at 500 mg/kg with resveratrol at 50 mg/kg had about 1.6 times more NAD+ in heart and 1.7 times more in muscle than mice given NMN alone (Bai et al., Pharmacology Research & Perspectives). Mouse doses and tissues do not translate directly to people, and we could not find a randomised human trial of NMN and resveratrol taken together.
So the honest answer depends on your goal:
- Raising NAD+ is the main aim: start with NMN. It has the clearer human data for that job.
- You are interested in the polyphenol itself: resveratrol has its own small human trials, including the memory study above.
- You want the combination most longevity routines use: take both, knowing the pair has not been trialled in people.
