- 80 healthy middle-aged adults took placebo or 300mg, 600mg or 900mg of NMN a day for 60 days. Blood NAD+ rose in every NMN group, highest at 600mg and 900mg, and six-minute walking distance improved more than with placebo (Yi et al., GeroScience, 2023).
- 108 older Japanese adults took 250mg of NMN or placebo once a day for 12 weeks. The group taking it in the afternoon showed the largest gains in a sit-to-stand test and the largest drop in drowsiness (Kim et al., Nutrients, 2022).
- A July 2026 pooled analysis of 15 randomised trials, at doses from 250mg to 2,000mg a day for up to 24 weeks, found no rise in adverse events or liver enzymes, and no clear effect on body weight, fasting glucose or blood fats (Yang et al., Nutrients, 2026).
Spermidine. The strongest results are in animals. Oral spermidine extended lifespan in mice and kept older mouse hearts working better (Eisenberg et al., Nature Medicine, 2016). In 829 adults aged 45 to 84 followed for about 20 years, higher spermidine intake from food was linked to lower death rates, which is an observational finding and cannot show cause (Kiechl et al., American Journal of Clinical Nutrition, 2018). In the one year-long capsule trial, 100 adults aged 60 to 90 took 0.9mg of spermidine a day or placebo for 12 months, and memory scores did not differ (Schwarz et al., JAMA Network Open, 2022).
Resveratrol. Results pull both ways. 46 healthy, overweight older adults who took 200mg a day for 26 weeks remembered more words than the placebo group (Witte et al., Journal of Neuroscience, 2014). But in 27 inactive men aged about 65 doing eight weeks of high-intensity training, 250mg a day of trans-resveratrol blunted the fitness and blood pressure gains seen with placebo (Gliemann et al., Journal of Physiology, 2013).
TMG. DoNotAge sells TMG to pair with NMN, and many people take the two together. We found no human trial showing that adding TMG changes what NMN does, so treat the pairing as a popular practice rather than a tested one.
Where any honest review should slow down is the jump from these findings to promises about human lifespan. The mouse data are strong. The human data are small, short or observational. Any brand, DoNotAge or ourselves included, that presents this as proof of longer human life is overselling it.
