The key point for dosing is that the NMN is fixed to the scoop. You cannot raise the NMN without raising the glycine, magnesium, vitamin C, vitamin D3 and everything else at the same rate. Is 500mg enough?

On the trial evidence, 500mg is a reasonable middle dose. Four of the trials above found changes at 250mg, and every trial above that measured blood NAD found that it rose. The one trial that compared doses side by side found the largest rise in blood NAD at 600mg and 900mg, with 300mg also clearly above placebo (Yi et al., GeroScience, 2023). None of these trials used exactly 500mg, so the best we can say is that 500mg falls between doses that raised blood NAD.

What “enough” means is also unclear. Most of these trials measured blood NAD, walking tests or questionnaires over 4 to 12 weeks. A higher blood NAD reading has not been shown to slow ageing, and no trial has tested a dose over years. The dose-comparison trial was funded by two companies, Aba Chemicals and Abinopharm, and its lead author works for Abinopharm, as the paper declares. That does not cancel the result, but it is a reason to treat “600mg worked best” as one finding, not a settled rule.

Can you take two scoops of AgeMate for a higher NMN dose?

AgeMate’s directions are one level scoop a day. Two scoops would give 1,000mg of NMN, which is above the highest dose in the dose-comparison trial (900mg) and below the 1,250mg used in the four-week safety study. It would also double everything else in the scoop:

  • glycine from 1.1g to 2.2g
  • magnesium citrate from 300mg to 600mg
  • vitamin C from 250mg to 500mg
  • zinc from 4mg to 8mg
  • vitamin D3 from 25mcg to 50mcg (2,000 IU)

On the single subscription, that is $5.26 a day. If you want more NMN rather than more of everything, a capsule lets you change the NMN on its own. Either way, talk to your GP or pharmacist before you go above a label dose, especially if you take other supplements that already contain magnesium, zinc or vitamin D.

Timing: when to take your daily 500mg

AgeMate’s directions put the scoop in the morning, and our NMN 500mg label also suggests the morning. That is a sensible habit, but the timing evidence is thin. The one trial designed to test timing split 108 older adults into morning and afternoon groups on 250mg a day for 12 weeks. The afternoon NMN group showed the largest improvements in a sit-to-stand test and in drowsiness, although some placebo groups improved too (Kim et al., Nutrients, 2022).

How long to take it

AgeMate’s label says to use it daily for up to 12 weeks unless a health professional advises otherwise. That lines up with the trials, which ran from 4 to 12 weeks, and with the lack of long-term data. Who should take care with NMN

The trials above reported few problems. Blood NAD rose without safety issues at up to 900mg a day over 60 days, and 1,250mg a day for four weeks caused no changes beyond normal variation in healthy adults (Fukamizu et al., Scientific Reports, 2022). The trials we cite ran for 12 weeks at most.

  • AgeMate’s label says adults only, notes that it contains phenylalanine and sulfites, and says it is not recommended for pregnant or breastfeeding women.
  • Do not take NMN if you are pregnant or breastfeeding, as there is no safety data for either group.
