Alpha-ketoglutarate (AKG) is a small molecule your cells make every day as part of the Krebs cycle, the chain of reactions that turns food into cellular energy. Ca-AKG is AKG bound to calcium, which makes a stable powder that fits in a capsule. It is also the form used in the research that made AKG popular as a healthy-ageing supplement.
Here is what that research shows, and what it doesn’t:
- Mice: at the Buck Institute, mice fed CaAKG lived longer, healthier lives with lower levels of inflammatory markers in their blood (Asadi Shahmirzadi and colleagues, 2020). Mouse results often don’t carry over to people.
- People, without a placebo group: a retrospective analysis of 42 self-reported healthy adults who took Rejuvant, a sustained-release product with 1 g of Ca-AKG a day plus vitamin A or D, found their DNA methylation age was an average of 8 years lower after 4 to 10 months (Demidenko and colleagues, 2021). There was no control group, the participants chose to buy the product, and one author sits on the board of the company that sells it. The authors themselves say a placebo-controlled trial is needed.
- People, with a placebo group: that trial, called ABLE, is running in Singapore. It enrolled 120 adults aged 40 to 60 and compares 1 g of sustained-release Ca-AKG with placebo for 6 months, with 3 months of follow-up (Sandalova and colleagues, 2023, Lim and colleagues, 2025). As of September 2026, its results on biological age haven’t been published.
So does AKG really work? The honest answer is that it looks promising in animals, and the only human data on biological age is uncontrolled. Anyone telling you Ca-AKG is proven to slow ageing in people is ahead of the evidence. If you take it, take it as a low-cost bet on a well-studied metabolite, not a sure thing.
