- NAD+ precursors (NMN and NR). Both raised NAD+ in blood in human trials. Whether that improves how mitochondria work in muscle is less clear. One NR trial that measured it directly found no change.
- Electron carriers (CoQ10 and methylene blue). CoQ10 is part of the chain your body already runs. Methylene blue can pick up and pass on electrons in cell studies.
- A Krebs cycle metabolite (Ca-AKG). Alpha-ketoglutarate is one of the cycle’s own intermediates. Most of its evidence comes from mice.

- Mitochondrial function didn’t. In 13 healthy adults with overweight, 1,000mg of NR a day for six weeks raised NAD+ markers in muscle, but had no effect on mitochondrial function, insulin sensitivity or energy metabolism (Remie et al., American Journal of Clinical Nutrition, 2020).

The human performance data are small. In a double-blind crossover trial, 17 healthy volunteers took 100mg or 300mg of CoQ10 a day, or a placebo, for eight days, then did a long, tiring cycling test. At 300mg, they held their sprint speed better and felt less fatigue than on placebo (Mizuno et al., Nutrition, 2008).
Dose against the trial: 150mg is half the 300mg dose that made a difference in that trial.
The label says it contains soya oil and not to take it while on warfarin without medical advice. Methylene blue can accept electrons and pass them to cytochrome c, a step in the energy chain. In human skin cells grown in the lab, very low concentrations raised complex IV activity by 30% and oxygen use by 37% to 70% (Atamna et al., FASEB Journal, 2008). That is a cell study, so it tells you how the molecule behaves, not what a capsule does in your body.
The main human trial gave 26 healthy adults aged 22 to 62 a single 280mg dose (about 4mg per kg) or a placebo, 13 people in each group. One hour later, brain scans showed more activity during attention and short-term memory tasks, and correct answers in memory retrieval rose 7% (Rodriguez et al., Radiology, 2016). That trial measured brain activity after one dose, not mitochondria.

Safety first: methylene blue must not be combined with SSRIs, SNRIs, MAOIs or other serotonergic medicines, because of the risk of serotonin toxicity. There was no placebo group, and one author sat on the board of the company selling the formula.
